the effects of follicular fluid on human x y bearing sperm ratio by fluorescent in situ hybridization (fish)

چکیده
background: some evidences showed that the secretion of uterine tube vagina and follicular fluid (ff) affects x and y-chromosome populations. sperm selection with x or y chromosome can added to oocyte for gender desired. the isolation of x y sperm have done and all efforts in this field are done to make culture media similar to in vivo condition. the objective of this study was to find if the ff can influence the ratio of x or y chromosomes therefore we added human ff to culture media to separate x and y sperms. materials and methods: normal semen sample from 36 healthy men were selected. then the samples were divided into control and experimental groups: control group sperms have been incubated with conventional culture media (ham's f-10) and experimental group with conventional culture media + 10% human ff. for sperm isolation swim up technique was used. after 24 hours of incubation slides smear were prepared. then we used the fluorescent in situ hybridization (fish) method to evaluate the effect of follicular fluid on the population ratio of x and y containing sperms. results: although the incubation of sperm in ff and ham's f-10 increased y sperm (59.44 % in control and 61.42% in experimental groups) in comparison with x sperm (40.56% in control and 38.5 in experimental groups) significantly (p

Introduction: To evaluate the 8 layer discontinues puresperm gradient in separation of human spermatozoa according to sex chromosomes by fluorescent in situ hybridization (FISH). Material and Methods: This study was carried out on three patient referring to Royan fertility and infertility center. Semen analysis was assessed according to WHO criteria. Each sample was divided into two aliquots, ...
متن کاملSperm processing affected ratio of X- and Y-bearing sperm
Studies showed that assisted reproductive technology changed the sex ratio at birth, but the effect of semen processing (swim-up technique and density-gradient separation) on enriching either Xor Y-bearing sperm is always controversial. We recruited 30 healthy males and their semen samples were obtained by masturbation after sexual abstinence for 2-7 days. Each sample was divided into 3 aliquot...
متن کاملThe Effect of Diffrent Factors on X- and Y Bearing Sperm in Bull Semen
Background: Newborn gender ratio is one of the most important factors in livestock industry. This ratio has affected on economic condition and controls sex-linked genetically transferred diseases. Offspring sex ratio is affected by such various factors. One of these factors is the Y to X bearing sperm ratio in normal semen that is called primary sex ratio, season and individual differences are ...
متن کاملFluorescent In Situ Hybridization (FISH) Fluorescent in situ hybridization (FISH) is a fluorescence probe
The importance of cancer biomarkers is now increasingly recognized and acted on by regulatory organizations, healthcare providers (from reimbursement through to practicing clinicians), cancer organizations, research institutes, and charities. For example, the American Joint Committee on Cancer (AJCC) states in the opening of its mission statement, “The purpose and scope of the AJCC is to create...
متن کاملTriple colour fluorescent in-situ hybridization for chromosomes X,Y and 1 on spare human embryos.
The potential for implantation of human embryos obtained by in-vitro fertilization is presumably determined to a large extent by their chromosomal constitution but cytogenetic analysis of preimplantation embryos has been hampered by a number of practical and technical problems. With the advent of fluorescent in-situ hybridization (FISH) a practical method for numerical chromosomal analysis has ...
